No cell walls, no new cancer cells

August 02, 2006

Cancer cells, like houses, need building materials for their walls. And as with a house, the cell wall needs to be built at just the right moment to protect and allow the construction of internal components. A team from the Uppsala Branch of the global Ludwig Institute for Cancer Research (LICR) has not only shown how the cell gets this timing right, but has also conducted proof-of-principle studies that indicate taking away the cell's bricks and mortar is a potential strategy for cancer control.

"New cells are created by the duplication of existing cells through a highly-organized process known as the cell cycle," explains lead author, Dr. Maite Bengoechea Alonso. "Last year we discovered that a protein called SREBP1 that regulates the synthesis of lipids needed for new cell walls was regulated during the cell cycle. Now we show that the SREBP1 protein actually controls the cell cycle."

Senior author, LICR's Dr. Johan Ericsson, realized that disrupting the function of SREBP1 might prevent the lipid synthesis required for new cell walls. "In fact, we literally stopped the cell cycle in its tracks by removing SREBP1 from cells. It seems that if you don't have SREBP1 activity, you can't make lipids, and if you don't have lipids, you can't make new cells."

According to Dr. Ericsson, who is also a Research Fellow of the Royal Swedish Academy of Sciences, this approach might one day form the basis of a new strategy for the long-term control of cancer. "Cancer cells divide uncontrollably, so their need for lipids is more urgent and continuous than normal cells. Treatment with an inhibitor of SREBP1 might reduce the rate of cancer cell proliferation to slow down tumor growth, or might enhance the effect of targeted therapies that aim to actually kill cancer cells."

New finding suggests prostate biopsy is not always necessary
Researchers at Wake Forest University School of Medicine and the University of Wisconsin-Madison have discovered that some elevated prostate-specific antigen (PSA) levels in men may be caused by a hormone normally occurring in the body, and are not necessarily a predictor of the need for a prostate biopsy.

Approved lymphoma drug shows promise in early tests against bone cancer
A drug already approved for the treatment of lymphoma may also slow the growth of the most deadly bone cancer in children and teens, according to an early-stage study published online today in the International Journal of Cancer.

Inhibitor of heat shock protein is a potential anticancer drug, Penn study finds
Like yoga for office drones, cells do have coping strategies for stress. Heat, lack of nutrients, oxygen radicals - all can wreak havoc on the delicate internal components of a cell, potentially damaging it beyond repair.

Alcohol activates cellular changes that make tumor cells spread
Alcohol consumption has long been linked to cancer and its spread, but the underlying mechanism has never been clear.

Chemosensitivity of cancer cells depends on their protein dependency
Two different anti-apoptotic proteins support cancer cell survival via an identical mechanism, yet differ in their sensitivity to chemotherapeutic drugs, report Brunelle et al. The study will be published online October 26, 2009 and in the November 2, 2009 print issue of the Journal of Cell Biology (JCB).

NEDD9 Protein Supports Growth of Aggressive Breast Cancer
Researchers at Fox Chase Cancer Center have demonstrated that a protein called NEDD9 may be required for some of the most aggressive forms of breast cancer to grow. Their findings, based on the study of a mouse model of breast cancer, are presented in a recent issue of Cancer Research, available on-line now.

One Renegade Cell: The Quest For The Origin Of Cancer (Science Masters)

One Renegade Cell: The Quest For The Origin Of Cancer (Science Masters)
by Robert A. Weinberg (Author)

For everyone whose life has been touched by cancer, One Renegade Cell tells the story of the search for the ultimate causes of this dreaded disease.

"One Renegade Cell…offers a breathtaking picture, both wonderful and frightening, of the fantastic intricacy of aberrant cellular functioning." -New York Times

"Part primer, part history and part meditation. [One Renegade Cell] succeeds on all counts." -Wall Street Journal

One of the leading cancer researchers in the world, Robert A. Weinberg is perfectly suited to describe the search for cancer's origins from the early days of this century to the present. Presuming little knowledge of biology, he tells how a cancer-causing virus was first discovered in 1909, how the correlation was made between chemical ...
