Changes in urine could lead to BSE test for live animals

September 08, 2008

Researchers have demonstrated that protein levels in urine samples can indicate both the presence and progress of Bovine Spongiform Encephalopathy (BSE) disease in cattle. Publishing their findings in BioMed Central's open access journal Proteome Science, the scientists hope that their discovery might lead to the development of a urine-based test that could prevent the precautionary slaughter of many animals as now occurs when the disease is detected.

The discovery of a new human variant of Creutzfeldt-Jakob Disease (vCJD), thought to be transmitted to humans via BSE-infected products, has resulted in many countries instituting extensive BSE testing programs for older animals. Diagnostic testing currently involves the detection of a misfolded "infectious" protein, or prion, in post-mortem brain tissue.

BSE has caused significant changes in the way beef products are produced and traded. If a simple and accurate urine test can be developed from this new knowledge, it could be performed on live animals and therefore may provide an alternative to current BSE surveillance procedures. It could also allow for the assessment of the health of breeding stock where post-mortem testing is not an option.

"We are hopeful that the knowledge that we've gained from this study will eventually lead to a live test," says Dr. David Knox, a researcher at the Public Health Agency of Canada's National Microbiology Laboratory in Winnipeg. "It may be possible to develop similar tests for other species as well, including humans with Creutzfeldt Jakob disease (CJD). A urine test for CJD could assist doctors to narrow down potential diagnoses for people with dementia".

Knox led a team of researchers who have demonstrated that the "protein profile" of cattle urine samples can indicate the presence of a BSE infection as well as how far the disease has advanced. The scientists analysed the proteins in urine samples taken from four infected and four healthy cows of the same age over the course of the disease. The proteins from the healthy and infected samples were compared using a technique called two-dimensional differential-gel electrophoresis (2-D DIGETM)

In these preliminary results a single protein was able to distinguish between those infected and control animals. In addition, the relative abundance of a set of proteins could accurately determine how far the disease had advanced.

"Our work shows that it is possible to identify biomarkers in urine that could be useful in the diagnosis and monitoring of disease progression in BSE and related transmissible spongiform encephalopathies," says Knox.

Pilot study relates phthalate exposure to less-masculine play by boys
A study of 145 preschool children reports, for the first time, that when the concentrations of two common phthalates in mothers' prenatal urine are elevated their sons are less likely to play with male-typical toys and games, such as trucks and play fighting.

Workplace BPA exposure increases risk of male sexual dysfunction
High levels of workplace exposure to Bisphenol-A may increase the risk of reduced sexual function in men, according to a Kaiser Permanente study appearing in the journal Human Reproduction, published by Oxford Journals.

Scientists take step toward simple and portable tuberculosis tests for developing world
Two billion people worldwide carry the pathogen that causes tuberculosis (TB), and most of them do not even know they are infected. This is because some 90 percent of people with TB have "latent" infections. They have no symptoms, they can't spread the disease to others and the bug remains dormant in their lungs -- often for years.

How much omega-3 fatty acid do we need to prevent cardiovascular disease?
A team of French scientists have found the dose of DHA (docosahexaenoic acid) that is "just right" for preventing cardiovascular disease in healthy men.

Chemicals in common consumer products may play a role in pre-term births
A new study of expectant mothers suggests that a group of common environmental contaminants called phthalates, which are present in many industrial and consumer products including everyday personal care items, may contribute to the country's alarming rise in premature births.

Description:
A novel and automated sequential injection procedure is proposed for the spectrophotometric determination of orthophosphate without requiring unstable chemical reducing species used in the classical molybdenum blue method. The flowing methodology is based on the on-line generation of the detectable species by electrochemical reduction of the 12-molybdophosphoric acid complex using a stainless steel tubular flow-through working electrode. The established method is linear up to 20mg/l P, with coefficients of...

Spectrum Labs Urine Luck Additive Formula 6.9

Spectrum Labs Urine Luck Additive Formula 6.9
by Spectrum Labs

Urine Luck is a chemical solution, which destroys nicotine metabolites in the urine to pass a pre-employment nicotine test. Current Urine Luck formulas (6.8 and above) are completely undetectable. The product consists of two tiny vials about the size of a pinkie finger. Pour both vials directly into the urine sample prior to submitting the sample for pre-employment nicotine testing. It is designed for a daily user because preparations are not needed, but should only be used on GC/MS tests. The Urine Luck changes the molecular structure of the compound, which also changes every chemical property of the compound. This is also why Urine Luck works better on nicotine tests than on pre-employment nicotine screens. Nicotine screens are an inexpensive and less precise way to detect Nicotine and...

Description:
This work describes an efficient, fast, and reliable analytical methodology for mercury determination in urine samples using stripping chronopotentiometry at gold film electrodes. The samples were sonicated in the presence of concentrated HC1 and H"2O"2 for 15min in order to disrupt the organic ligands and release the mercury. Thirty samples can be treated over the optimized region of the ultrasonic bath. This sample preparation was enough to allow the accurate stripping chronopotentiometric determination of...
