Circulating Nucleic Acids in Plasma or Serum (Annals of the New York Academy of Sciences)
by International Symposium on Circulating Nucleic Acids in Plasma (Author), Philippe Anker (Editor), Maurice Stroun (Editor), Serum 1 (Editor)
Free DNA present in both healthy and diseased human plasma has been found to express specific point mutations that may serve as diagnostic signposts - perhaps to reliable, non-invasive tests for breast and lung cancer, haematopoetic malignancies, colorectal or pancreatic carcinoma, and other tumours. In another research area, foetal DNA has been detected in maternal serum which presents an early-horizon alternative in utero test for gender. A parallel discovery of rearranged immunoglobin heavy-chain DNA in the plasma of patients wit Hodgkin's disease or acute B cell leukaemia has also emerged. These fast-emerging indicators suggest that it is time to standardize research techniques and develop protocols that can take a measure of circulating DNA's usefulness as a diagnostic tool. This...

Description: Glycidamide (GA)-induced mutagenesis in mammalian cells is not very well understood. Here, we investigated mutagenicity and DNA repair of GA-induced adducts utilizing Chinese hamster cell lines deficient in base excision repair (BER), nucleotide excision repair (NER) or homologous recombination (HR) in comparison to parent wild-type cells. We used the DRAG assay in order to map pathways involved in the repair of GA-induced DNA lesions. This assay utilizes the principle that a...

Iron chelators reduce chromosomal breaks in ataxia-telangiectasia cells [An article from: DNA Repair]
by R.E. Shackelford (Author), Y. Fu (Author), R.P. Manuszak (Author), T. Brooks (Author)
This digital document is a journal article from DNA Repair, published by Elsevier in 2006. The article is delivered in HTML format and is available in your Amazon.com Media Library immediately after purchase. You can view it with any web browser.
Description: Ataxia-telangiectasia (A-T) is characterized by ataxia, genomic instability, and increased cancer incidence. Previously, iron chelator concentrations which suppressed normal cell colony formation increased A-T cell colony formation. Similarly, iron chelators preferentially increased A-T cell colony formation following peroxide exposure compared to normal cells. Last, A-T cells exhibited increased short-term sensitivity to labile iron exposure compared to normal cells, an event corrected by recombinant ATM (rATM)...

Nutty Professor II - The Klumps (Collector's Edition)
Starring: Eddie Murphy, Janet Jackson, Nikki Cox, Chris Elliott, Enya Flack
America's funniest family back for seconds! Eddie Murphy is hilarious when he stars as the entire Klump family in this enormous comedy blockbuster. The hilarity begins when professor Sherman Klump finds romance with fellow DNA specialist, Denise Gaines (Janet Jackson), and discovers a brilliant formula that reverses aging. But Sherman's thin and obnoxious alter ego, Buddy Love, wants out… and a big piece of the action. And when Buddy gets loose, things get seriously nutty. Now, it's up to Mama, Papa, Ernie and Granny Klump to throw their weight around and save the day in this whopping, gut-busting comedy Good Morning America calls an "incredible piece of filmmaking." Applauded by audiences and critics alike, Roger Ebert and Richard Roeper give Nutty Professor II: The Klumps: "two thumbs...

DNA polymerases @h and @k are responsible for error-free translesion DNA synthesis activity over a cis-syn thymine dimer in Xenopus laevis oocyte extracts [An article from: DNA Repair]
by Y. Yagi (Author), D. Ogawara (Author), S. Iwai (Author), F. Hanaoka (Author), Akiyama (Author)
This digital document is a journal article from DNA Repair, published by Elsevier in . The article is delivered in HTML format and is available in your Amazon.com Media Library immediately after purchase. You can view it with any web browser.
Description: In translesion synthesis (TLS), specialized DNA polymerases (pols) facilitate progression of replication forks stalled by DNA damage. Although multiple TLS pols have been identified in eukaryotes, little is known about endogenous TLS pols and their relative contributions to TLS in vivo because of their low cellular abundance. Taking advantage of Xenopus laevis oocyte cells, with their extraordinary size and abundant enzymes involved in DNA metabolism, we have identified and characterized endogenous TLS pols for DNA damage...

Mechanism of interaction between human 8-oxoguanine-DNA glycosylase and AP endonuclease [An article from: DNA Repair]
by V.S. Sidorenko (Author), G.A. Nevinsky (Author), D.O. Zharkov (Author)
This digital document is a journal article from DNA Repair, published by Elsevier in 2007. The article is delivered in HTML format and is available in your Amazon.com Media Library immediately after purchase. You can view it with any web browser.
Description: Human 8-oxoguanine-DNA glycosylase (OGG1) is the main human base excision protein that removes a mutagenic lesion 8-oxoguanine (8-oxoG) from DNA. Since OGG1 has DNA glycosylase and weak abasic site (AP) lyase activities and is characterized by slow product release, turnover of the enzyme acting alone is low. Recently it was shown that human AP endonuclease (APE1) enhances the activity of OGG1. This enhancement was proposed to be passive, resulting from APE1 binding to or cleavage of AP sites after OGG1 dissociation. Here...
