First human data presentation: Melina E. Marmarelis, MD, will present the ongoing Phase 1 program in a Rapid Oral session on October 25 in Madrid.
Mesothelioma focus: The investigational oral pan-TEAD inhibitor target the Hippo/TEAD pathway, which is implicated in cancer biology and is conducting multicenter Phase 1 study studies in mesothelioma and other advanced solid tumors.
AI-enabled development: ISM6331 was designed and optimised using Insilico’s generative chemistry engine, Chemistry42. It is one of two oncology pipelines wholly owned by the company and advanced through clinical trials. .
CAMBRIDGE, Mass., October 21, 2026 — Insilico Medicine ("Insilico", HKEX: 3696), a clinical-stage drug discovery company powered by generative artificial intelligence (AI), today announced that the abstract of first-in-human multicenter Phase 1 study of ISM6331, an AI-designed pan-TEAD inhibitor, in patients with mesothelioma or other advanced solid tumors, has been published on the official website of the 2026 European Society for Medical Oncology (ESMO). Detailed and updated data from the study will be presented during the ESMO “Rapid Oral Presentation” session on October 25, 2026.
The presentation will address the clinical evaluation of ISM6331 in patients with mesothelioma or other advanced solid tumors. It provides an opportunity for the oncology community to examine the development of an AI-designed molecule targeting the Hippo/TEAD pathway.
Mesothelioma is a rare and aggressive cancer for which researchers continue to investigate additional treatment approaches, including for patients whose disease progresses after treatment. TEAD transcription factors participate in signaling downstream of the Hippo pathway, which helps regulate cell growth and tissue maintenance. Dysregulation of this signaling network can contribute to cancer biology, providing a rationale for investigating TEAD inhibition in selected tumors.
The ongoing multicenter, open-label Phase 1 study evaluates the safety, tolerability, pharmacokinetics and preliminary antitumor activity of ISM6331. Study objectives include assessing dose-limiting toxicities and determining a recommended Phase 2 dose. Data from the study have been submitted for presentation at the ESMO Congress 2026 in Madrid, Spain, on October 25, 2026, and will be discussed in a subsequent announcement.
“Clinical development is where a molecule’s scientific hypothesis is tested against the needs and complexity of patient care,” said Halle Zhang, PhD, Vice President, Clinical Development – Oncology at Insilico Medicine. “With ISM6331, our focus is on understanding the compound’s safety, identifying an appropriate dose and evaluating its preliminary antitumor activity in patients with mesothelioma and other advanced solid tumors. We look forward to discussing the program with the oncology community at ESMO.”
Insilico used its Chemistry42 generative chemistry platform to support the design of ISM6331. The program combines computational molecular design with laboratory evaluation and clinical development. Its progression illustrates how an AI-designed candidate can move into human testing, while the evidence needed to establish clinical benefit remains a separate question for clinical investigation.
Insilico nominated ISM6331 as a preclinical candidate in June 2023, and the program entered a global multicenter Phase 1 study in January 2025. The company has publicly announced U.S. FDA Orphan Drug Designation for mesothelioma and Fast Track Designation for adult patients with previously treated, unresectable malignant pleural mesothelioma . These designations support development and do not establish clinical efficacy or constitute marketing approval.
The program forms part of Insilico’s broader drug discovery and development portfolio. Leveraging its generative AI platform, Insilico has continuously demonstrated scalable and reproducible R&D efficiency. Since 2021, the company has nominated 35 developmental candidates, with 13 of them receiving IND clearance or approval. Notably, Insilico broke its annual pipeline output record by nominating 11 PCCs in just the first 10 months of 2026. Among these milestones, Rentosertib has advanced into Phase III clinical trials for idiopathic pulmonary fibrosis (IPF), taking a crucial step toward clinical validation and commercialization.
